This study presents a comprehensive molecular and biochemical characterization of bacteria isolated from livestock soil environments within the Oyo State College of Agriculture and Technology (OYSCATECH), Igboora, focusing on cattle kraal and pig house soils. Using standard serial dilution, Gram staining, biochemical tests, DNA extraction, PCR, and sequencing, a wide diversity of Gram-positive and Gram-negative bacteria were identified across multiple genera, including Bacillus, Staphylococcus, Pseudomonas, Escherichia, Salmonella, Neisseria, Streptococcus, and Enterococcus. Molecular analysis revealed close phylogenetic relationships between selected isolates and reference strains in the NCBI GenBank, notably Pseudomonas pseudoalcaligenes, Escherichia coli, and Salmonella enterica. The findings highlight livestock soil as a reservoir of bacteria with dual implications—some with pathogenic potential that may threaten animal and public health, and others with possible protective or beneficial roles. The study underscores the importance of molecular surveillance, improved sanitary practices in livestock housing, and further research aimed at harnessing beneficial soil bacteria while mitigating disease transmission risks.
